The audience laughs when Iago says he would rather lose his tongue than say something bad about Cassio because all Iago does is say bad things about others, including Cassio.

Such a statement is ridiculous to say because Cassio has just gotten into a fight with Montano because of things Iago said. Iago is clearly a man of no scruples and a liar.
